Estrogen biology depends on genes that make estrogens, receive estrogen signals, process estrogen metabolites, and help clear them from the body. A DNA file cannot measure hormone levels, but it can show variants in genes that are part of those pathways.

CYP19A1

CYP19A1 encodes aromatase, the enzyme that converts androgens into estrogens. Variants in or near this gene can be relevant when reviewing estrogen production and hormone-sensitive traits.

CYP17A1

CYP17A1 helps produce steroid hormone precursors. It sits upstream of estrogen production, so it is often reviewed with other steroid metabolism genes.

Receptor signaling

ESR1 and ESR2

ESR1 and ESR2 encode estrogen receptors. These receptors bind estrogen and affect gene expression in target tissues. Variants in receptor genes may be reviewed when the question is hormone sensitivity rather than hormone production.

Metabolism

CYP1A1, CYP1A2, CYP1B1, and CYP3A4

Several cytochrome P450 genes process estrogen metabolites. CYP1A1 and CYP1A2 participate in 2-hydroxylation. CYP1B1 participates in 4-hydroxylation. CYP3A4 contributes to broader steroid metabolism.

COMT

COMT helps methylate catechol estrogens. This step changes reactive estrogen metabolites into forms that are easier to clear.

Elimination and recycling

SULT1E1 and UGT1A1

SULT1E1 and UGT1A1 help attach chemical groups to estrogen metabolites. This usually makes the metabolites more water-soluble and easier to eliminate.

STS

STS can convert sulfated, less active estrogen forms back into active forms. This is one reason estrogen metabolism is better reviewed as a pathway than as one gene at a time.

Then review variant consequence, allele frequency, known clinical annotations, and whether the variant fits the question you are asking. A variant in an estrogen-related gene does not by itself explain hormone symptoms or disease risk.

Use genetic findings as context for a healthcare discussion, especially when the question involves symptoms, medication, cancer risk, fertility, or hormone treatment.
